本研究では、HPLC-PDA/MS/MSを用いたL. ()の化学的プロファイリングと、ヒト乳癌細胞株(MCF-7)、ヒト大腸癌細胞(HCT-116)およびヒト肝細胞癌細胞株(Huh-7)に対する細胞毒性の評価を行った。生存率は、中性赤色取り込みアッセイにより決定した。この研究により、37種類の二次代謝物が同定された。主要な9種類の化合物を仮想ドッキングさせ、MST3リガンドタンパク質の3次元結晶構造を用いて、アポトーシスやがん細胞増殖を制御することで、腫瘍増殖抑制に果たす役割を決定した。サンバコシドAとモリハウスシドCの2つの化合物は、MST3タンパク質に対して-9.91, -9.57 kcal/molの高親和性を示し、吸収・分布・代謝・排泄・毒性予測(ADMET)を行った結果、両化合物とも変異原性、腫瘍原性、非刺激性の作用はなく、細胞毒性化合物の有益な供給源として利用できることがわかった。
In this study chemical profiling of L. () using HPLC-PDA/MS/MS and evaluation of its cytotoxicity towards the human breast cancer cell line (MCF-7), human colorectal cancer cell (HCT-116) and human hepatocellular carcinoma (Huh-7) cell lines. The viability % was determined by the neutral red uptake assay. The study led to the identification of 37 secondary metabolite; major nine compounds were subjected to virtual docking to determine their role in tumour growth inhibition by controlling apoptosis and cancer cell proliferation using the 3D crystal structure of MST3 ligand protein. Two compounds; sambacoside A and molihauside C, showed high-affinity values of (-9.91, -9.57) kcal/mol against MST3 protein prediction of absorption, distribution, metabolism, excretion and toxicity (ADMET) was performed and revealed no mutagenicity, no tumorigenicity and non-irritant actions of both compounds, so could be used as a beneficial source for cytotoxic compounds.
