Clostridium botulinum neurotoxins (BoNTs) は、運動ニューロンからのアセチルコリン放出を阻害することで弛緩性麻痺を引き起こすが、微量であればこの特性が治療薬として利用される。BoNTは、神経細胞膜を標的とする結合ドメイン(H )、転座ドメイン(H )、触媒ドメイン(LC )の3つのドメインから構成されています。ここでは、BoNTのサブタイプである/A5(H /A5)と/A6(H /A6)の結合ドメインの高分解能結晶構造を示す。これらの構造は、他のサブタイプで同定されたコアフォールディングが維持されていることを示しているが、期待される受容体結合部位に微妙な違いがあることを示している。
Clostridium botulinum neurotoxins (BoNTs) cause flaccid paralysis through inhibition of acetylcholine release from motor neurons; however, at tiny doses, this property is exploited for use as a therapeutic. Each member of the BoNT family of proteins consists of three distinct domains: a binding domain that targets neuronal cell membranes (H ), a translocation domain (H ), and a catalytic domain (LC). Here we present high-resolution crystal structures of the binding domains of BoNT subtypes /A5 (H /A5) and /A6 (H /A6). These structures show that the core fold identified in other subtypes is maintained, but with subtle differences at the expected receptor binding sites.
