ヒトにおける侵襲性アスペルギルス症の原因菌として最も一般的な菌種に属する30株(, , , ,および)に対するイサブコナゾールと免疫抑制剤(タクロリムス,シクロスポリンA,シロリムス)との相互作用を,EUCAST基準抗真菌感受性試験法に基づいたマイクロダイリューションチェッカーボード法によりin vitroで評価した。結果の解釈は分画阻害濃度指数に基づいて行った。isavuconazoleとtacrolimus,cyclosporin A,sirolimusの併用は,それぞれ56株,20株,10%の分離株に対して相乗効果を示した。興味深いことに,イサブコナゾールとタクロリムスの併用による相乗効果は,大多数のアゾール耐性株と,イサブコナゾールの最小阻害濃度が8μg/mL以上のすべての分離株で認められた。試験したどの組み合わせにおいても、拮抗的相互作用は観察されませんでした。
The interaction of isavuconazole with immunosuppressors (tacrolimus, cyclosporin A, or sirolimus) against 30 isolates belonging to the most common species responsible for invasive aspergillosis in humans (, , , , and ) was evaluated in vitro by a microdilution checkerboard technique based on the EUCAST reference method for antifungal susceptibility testing. The interpretation of the results was performed based on the fractional inhibitory concentration index. The combination of isavuconazole with tacrolimus, cyclosporin A, or sirolimus, was synergistic for 56, 20, or 10% of the isolates, respectively. Interestingly synergy of the combination of isavuconazole with tacrolimus was also achieved for the majority of azole-resistant isolates of , and for all isolates with isavuconazole minimal inhibitory concentrations ≥ 8 µg/mL. Antagonistic interactions were never observed for any combination tested.
