内因性の生理活性脂質は、健康や病気に関与する細胞や分子のプロセスの多くを調節する複雑なネットワークの一部であり、その中でも炎症は最も顕著な例の一つです。炎症は、化学的、機械的、または微生物による損傷を受けた場合に引き金となり、損傷の原因を根絶し、組織機能を回復させることを目的とした、保存された防御機構として機能しています。しかしながら、過剰な炎症性シグナル、またはプロ分解/抗炎症経路の障害は、慢性炎症を引き起こし、慢性疾患の特徴となっています。内因性の生理活性脂質のすべての主要なクラス - すなわち、エイコサノイド、特殊なプロ分解脂質メディエーター、リゾグリセロホップ脂質、エンドカンナビノイド - は、癌、糖尿病、アテローム性動脈硬化症、喘息、自己免疫疾患や神経変性疾患、炎症性腸疾患などの疾患を特徴づける慢性炎症に一貫して関与しています。このレビューでは、慢性炎症性疾患の発症過程における内因性生理活性脂質の関与に関する現在の知見を収集しています。
Endogenous bioactive lipids are part of a complex network that modulates a plethora of cellular and molecular processes involved in health and disease, of which inflammation represents one of the most prominent examples. Inflammation serves as a well-conserved defence mechanism, triggered in the event of chemical, mechanical or microbial damage, that is meant to eradicate the source of damage and restore tissue function. However, excessive inflammatory signals, or impairment of pro-resolving/anti-inflammatory pathways leads to chronic inflammation, which is a hallmark of chronic pathologies. All main classes of endogenous bioactive lipids - namely eicosanoids, specialized pro-resolving lipid mediators, lysoglycerophopsholipids and endocannabinoids - have been consistently involved in the chronic inflammation that characterises pathologies such as cancer, diabetes, atherosclerosis, asthma, as well as autoimmune and neurodegenerative disorders and inflammatory bowel diseases. This review gathers the current knowledge concerning the involvement of endogenous bioactive lipids in the pathogenic processes of chronic inflammatory pathologies.
