ほとんどの浸透圧ストレスは植物の成長と発達に有害であるが、特定の干ばつやポリエチレングリコール(PEG)によって誘発された軽度の浸透圧ストレスは植物の根の成長を促進する。この応答の基礎となる制御機構は未だ解明されていない。ここでは、GLYCOGEN SYNTHASE KINASE 3 (GSK3)遺伝子()と()が .エチレングリコール(PEG)の軽度の浸透圧ストレス(-0.4 MPa)応答に関与していることを報告する。浸透圧ストレスを模擬した異なる濃度の PEG を注入した培地で生育させた場合、野生型(WT)及び植物は、軽度の浸透圧ストレス(-0.4 MPa)では根の生長が促進されたが、比較的強い浸透圧ストレス(-0.5, -0.6, -0.7 MPa)では根の生長が抑制された(-0.5, -0.6, -0.7 MPa)。また、-0.4MPa処理ではWTよりも根の生長刺激に敏感であり、軽度のストレスによる根の生長応答を阻害していることが示唆された。また、WTと3つの水ポテンシャル(-0.25 MPa, -0.4 MPa, -0.6 MPa)下の植物のRNA-seq解析を行った結果、細胞壁恒常性に関与する10の異なる発現候補遺伝子が、軽度ストレス条件(-0.4 MPa)に応答して制御され、根の成長を制御していることが明らかになった。また、プロモーターモチーフと転写因子結合解析の結果、基本ヘリックス(bHLH)転写因子bHLH69/LJRHL1-LIKE2(LRL2)が、ほとんどの-0.4MPa応答性遺伝子の発現を直接制御している可能性が示唆された。これらの知見は、軽度の浸透圧ストレス(-0.4MPa)が植物の成長を促進し、GSK3ファミリーキナーゼ遺伝子と軽度の浸透圧ストレスに応答して根の成長誘導に負の役割を果たしていることを示している。
Although most osmotic stresses are harmful to plant growth and development, certain drought- or polyethylene glycol (PEG)-induced mild osmotic stresses promote plant root growth. The underlying regulatory mechanisms of this response remain elusive. Here, we report that the GLYCOGEN SYNTHASE KINASE 3 (GSK3) genes () ) and () are involved in the mild osmotic stress (-0.4 MPa) response in . When grown on plant medium infused with different concentrations of PEG to mimic osmotic stress, both wild-type (WT) and plants showed stimulated root growth under mild osmotic stress (-0.4 MPa) but repressed root growth under relatively strong osmotic stress (-0.5, -0.6, -0.7 MPa) as compared to the mock condition (-0.25 MPa). The root growth stimulation of was more sensitive to -0.4 MPa treatment than was that of WT, indicating that and inhibit the mild stress-induced root growth response. RNA-seq analysis of WT and plants under three water potentials (-0.25 MPa, -0.4 MPa, -0.6 MPa) revealed 10 differentially expressed candidate genes mainly involved in cell wall homeostasis, which were regulated by and to regulate root growth in response to the mild stress condition (-0.4 MPa). Promoter motif and transcription factor binding analyses suggested that the basic helix-loop-helix (bHLH) transcription factor bHLH69/LJRHL1-LIKE 2 (LRL2) may directly regulate the expression of most -0.4 MPa-responsive genes. These findings indicate that mild osmotic stress (-0.4 MPa) promotes plant growth and that the GSK3 family kinase genes and play a negative role in the induction of root growth in response to mild osmotic stress.
