高温環境下でのキシラナーゼを用いたキシランから糖への効率的な生分解反応

超好熱性キシラナーゼは、バイオマス利用の過程でキシランから糖へのバイオコンバージョンに重要な役割を果たしている。このような耐熱性キシラナーゼの需要の高まりに対応するためには、指向性進化に基づく新規キシラナーゼの発見や既存のキシラナーゼの改良が有効なアプローチとなることが期待されています。本研究では、Thermotoga neapolitana(Tne)由来のキシラナーゼB遺伝子(CTN_0623)をクローニングし、ヒートショックベクターpHshを用いて大腸菌にキシラナーゼB(以下、TnexlnB)を多量に可溶化発現させた。TnexlnBは、75℃、pH6.0で最も高いエンド-β-1,4-キシランヒドロラーゼ活性を示した。さらに、1mMMg, Ba, Caを添加することで、TnexlnBの活性をそれぞれ31%, 37%, 53%向上させることができた。また、TnexlnBの最後の3つのらせんに部位特異的に変異を加えることで、最適温度は85℃に達した。ワイドタイプと最良変異体の両方について、シラカバのキシランに対するKとVを以下のように決定した。1.09mg/mL、191.76U/mgおよび0.29mg/mL、376.42U/mgであった。さらに、この変異体は熱安定性に優れており(90℃で1時間後には酵素活性の80%以上を維持)、高温でのバイオマス分解に適した酵素であることが明らかになった。
Hyperthermophilic xylanases play a critical role in bioconversion from xylan to sugar in the process of biomass utilization. The discovery of new or improvement of existing xylanases based on directed evolution is expected to be an effective approach to meet the increasing demand of thermostable xylanases. In this work, a xylanase B gene (CTN_0623) from Thermotoga neapolitana (Tne) was cloned and xylanase B (herein named TnexlnB) was solubly expressed in E. coli with a high amount using a heat shock vector pHsh. TnexlnB showed the highest endo-β-1,4-xylan hydrolase activity at 75 °C and pH 6.0. Additionally, 1 mM Mg, Ba and Ca improved the activity of TnexlnB by 31%, 37%, and 53%, respectively. The optimal temperature reached 85 °C by site-directed mutation at the last three helices of TnexlnB. K and V towards birchwood xylan were determined for both wide type and the best mutant, as follow: 1.09 mg/mL, 191.76 U/mg and 0.29 mg/mL, 376.42 U/mg, respectively. Further characterization highlighted good thermal stability (>80% of enzymatic activity after 1 h at 90 °C) for the best mutant, which made this enzyme suitable for biomass degradation at high temperature.
