ダプトマイシンは、土壌ブドウ球菌Streptomyces roseosporus由来のカルシウム依存性環状リポペプチドであり、その抗生物質的性質から薬剤耐性グラム陽性感染症の治療薬として重要な役割を果たしています。臨床的には、グラム陽性皮膚・皮膚構造物感染症（SSSI）、黄色ブドウ球菌性菌血症、メチシリン耐性黄色ブドウ球菌（MRSA）を含む黄色ブドウ球菌に関連した右側心内膜炎の治療に最も一般的に使用されています。また、腸球菌感染症にも「適応外」で使用されています。この臨床的に重要な抗菌薬の作用機序（まだ完全には解明されていないが），耐性機序，生合成に関する研究は非常に多く行われている。我々は後者の側面を詳細にカバーしていますが、このレビューの主な焦点は、この重要なクラスのリポペプチド抗生物質の構造-活性-毒性に関する最も包括的で最新のリファレンスを提供することです。

Daptomycin is a calcium-dependent cyclic lipodepsipeptide derived from the soil saprotroph Streptomyces roseosporus and its antibiotic properties make it a key agent for treatment of drug-resistant Gram-positive infections. It is most commonly used clinically for the treatment of Gram-positive skin and skin structure infections (SSSI), Staphylococcus aureus bacteraemia, and right-sided endocarditis infections associated with S. aureus, including methicillin resistant S. aureus (MRSA). It has also been used 'off-label' for Enterococcal infections. There has been a tremendous amount of research investigating its mode of action (which has yet to be fully elucidated), resistance mechanisms and biosynthesis of this clinically important antimicrobial agent. Although we cover the latter aspects in detail, the primary focus of this review is to provide the most comprehensive and up-to-date reference for the medicinal chemist on the structure-activity-toxicity of this important class of lipopeptide antibiotics.
