本研究では、広く使用されている農薬であるエトプロホス(Etho)と重金属カドミウム(Cd)を単回及び反復投与した場合の雄性マウスの血液学的、生化学的、生殖学的及び細胞遺伝学的パラメータに及ぼすリスク評価方法を検討した。その結果、被験有害物質(Etho及びCd)の亜致死量(1/50 LD)では、体重及び臓器重量、血液学的プロフィール、アセチルコリンエステラーゼ(AChE)活性が低下することが明らかになった。被験汚染物質は、アスパラギン酸アミノトランスフェラーゼ(AST)およびアラニンアミノトランスフェラーゼ(ALT)を含む肝機能のパラメータ、ならびにクレアチニンおよび尿素を含む腎機能検査のパラメータを有意に増加させた。また、精子異常数(SA)や染色体異常数(CA)や有糸分裂指数(MI)を増加させることで細胞遺伝学的検査を刺激し、生殖機能検査にも悪影響を及ぼす。この探索的研究の全体的な結果は、試験した2つの化合物(EthoおよびCd)の共存は、電池で測定されたすべてのバイオマーカー、特に反復処理(14回投与)において、それぞれの化合物単独の場合よりも顕著な効果を引き起こす傾向があったことを示唆している。また、エンドポイントとして様々な簡易で感度の高いアッセイを組み合わせることで、包括的な情報が得られ、他の一般的に遭遇する環境汚染物質の潜在的な影響を評価するためのより良い洞察が得られました。
This work aimed to study the risk assessment procedures of a combination of single and repeated dose of the widely used pesticide, ethoprophos (Etho) and heavy metal cadmium (Cd), on the hematological, biochemical, reproduction and cytogenetic parameters in male mice. The results revealed that the sub-lethal dose (1/50 LD) of the tested toxic substances (Etho and/or Cd) reduced the body and organ weights, the most hematological profile and the activity of acetylcholine esterase (AChE). The tested pollutants significantly increased the parameters of liver function, including aspartate aminotransferase (AST) and alanine aminotransferase (ALT), as well as renal function tests, including creatinine and urea. In addition, they have deleterious effects on reproductive function tests by stimulating the number of sperm abnormalities (SA) and cytogenetic assays by increasing the frequency of chromosomal abnormalities (CA) and the mitotic index (MI). The overall results of this exploratory study suggest that the co-existence of the two tested compounds (Etho and Cd) had the propensity to cause a more pronounced effect than that of each compound alone on all the battery measured biomarkers, especially in the repeated treatment (14 doses) than that in the single one. Also, the combination of a range of simple and sensitive assays as endpoints gives a comprehensive picture and provides better insights to evaluate the potential effects of other commonly encountered environmental pollutants.
